In the final episode of our teach-in series, CEO Nigel Theobald introduces N4 101, our lead RNA-based therapeutic for Inflammatory Bowel Disease (IBD). This orally delivered treatment is designed to target macrophage cells, the key drivers of gut inflammation, by combining two RNA therapies on a single Nuvec® nanoparticle—a unique capability of our delivery platform.

N4 101 works through a dual-action approach:

siRNA knocks down TNF-alpha, reducing the body’s overproduction of this inflammatory agent.

mRNA boosts IL-10, a naturally occurring anti-inflammatory, helping the body fight back.

A mannose ligand is attached to enhance uptake into macrophage cells, ensuring more effective targeting.

Nigel also shares the latest progress, including successful in-vitro testing, demonstrating that N4 101 can simultaneously reduce TNF-alpha and increase IL-10 production. Next steps include optimising RNA loadings before moving to IBD mouse model testing to assess its impact on inflammation reduction.
