As RNA-based medicines rapidly advance, from vaccines to next-generation therapies for cancer and chronic disease, one truth remains clear: these powerful molecules are only as effective as the systems that deliver them.

The promise of RNA—and the problem holding it back

RNA therapeutics represent one of the most promising drug classes of the last decade. By mimicking or modifying the body’s own RNA, these treatments can:

Replace or silence faulty gene expression,

Target previously “undruggable” proteins,

And potentially even cure rather than manage chronic conditions.

However, RNA therapies face one stubborn barrier: delivery.

RNA is inherently unstable,it degrades rapidly in the bodyand it struggles to cross cell membranes. It rarely reaches the right cells—and when it does, often fails to escape the endosome to carry out its function.

Most current delivery systems—such as lipid nanoparticles (LNPs) or viral vectors—are limited in scope:

LNPs are largely restricted to the liver.

Viral vectors carry safety risks and manufacturing complexities.

Both typically rely on injection and often require high doses to achieve therapeutic effect.

These limitations restrict the types of diseases RNA therapies can treat—and the patients who can benefit from them.

N4 Pharma’s Nuvec® delivery system is designed to overcome these barriers.

Nuvec is a silica nanoparticle with a proprietary spiky surface structure that physically traps RNA and other nucleotides, protecting them from degradation and enabling controlled delivery into target cells.

This dual approach—delivered via a pH-controlled enteric capsule—is designed to reduce inflammation at the source, without systemic side effects or injection.

In preclinical studies, N4-101 has demonstrated:

Elevated IL-10 levels in the gut and bloodstream

These findings underscore Nuvec’s potential not just for IBD, but for a wide range of localized, targeted RNA applications.
